PUMA (p53 Up-Regulated Modulator of Apoptosis) is a pro-apoptotic protein belonging to the BH3-only subgroup of the BCL-2 protein family. It plays a critical role in mediating apoptosis in response to various cellular stresses by antagonizing anti-apoptotic BCL-2 family members, ultimately leading to mitochondrial outer membrane permeabilization and caspase activation. It is transcriptionally regulated by p53 and other factors, linking it to cancer development and therapy resistance.
Binds to and antagonizes anti-apoptotic BCL-2 family members, promoting BAX/BAK activation and mitochondrial outer membrane permeabilization.
