Overview

Paired box protein Pax-2 (PAX2) is a transcription factor of the paired box gene family, defined by a conserved DNA-binding paired box domain[1][2]. PAX2 plays key roles in embryonic development of the eyes, ears, brain, spinal cord, kidney, urinary tract, and genital tract, and is involved in tissue patterning and cellular differentiation[2][1][5]. After birth, PAX2 continues to function in cell survival and epithelial maintenance[2][5]. It acts as both a transcriptional activator and repressor, controlling genes involved in organogenesis and cell proliferation, most notably cyclin D1 in cancer cells[4]. Mutations in PAX2 cause renal coloboma syndrome and other congenital anomalies, while its dysregulation is implicated in various cancers where it promotes tumor progression and vasculogenic mimicry[5][4][3]. There are four main isoforms generated by alternative splicing[3]. PAX2 does not have approved targeted therapies, but is an active area of research in cancer and developmental biology.
