Overview

Paired box protein Pax-9 (PAX9) is a transcription factor from the paired box gene family that plays a critical role in mammalian development, particularly in the formation of teeth and craniofacial structures[1][2][3][4][5]. PAX9 binds to DNA using its paired domain and regulates the expression of genes necessary for odontogenesis, ribosome biogenesis, and other developmental pathways[1][2]. In humans, mutations in PAX9 are a common cause of familial tooth agenesis (oligodontia), usually affecting the posterior teeth without other systemic abnormalities[2][3][4][5]. PAX9 has also been implicated in the pathogenesis of certain cancers such as lung cancer, where gene amplification may contribute to oncogenesis[3]. There are currently no drugs known that target PAX9 directly, and it is not commonly considered a therapeutic pharmacological target, but it is an important biomarker and gene of interest for developmental biology and genetic disease studies[2][3][4][5].
