Paired-like homeodomain transcription factor LEUTX (LEUTX) is a PRD-like homeobox gene encoding a transcription factor that is expressed almost exclusively in human embryos during preimplantation development, specifically between the 4- and 8-cell stages[3][2][7]. LEUTX plays a crucial role in embryo genome activation by acting as a transcriptional activator of numerous genes necessary for the transition from maternal- to embryonic-controlled gene expression[2][3][4]. It binds to a specific DNA motif (TAATCC), and through interactions with histone acetyltransferases such as EP300 and CBP, via its 9 amino acid transactivation domain (9aaTAD), it enhances chromatin accessibility and drives expression of downstream targets[2]. LEUTX is essential for initiating the gene regulatory cascades in early human development, but after the 8-cell stage, its expression is silenced and it is not known to function in adult tissues. There is no evidence that LEUTX is a therapeutic target or that it is involved in disease states outside of its critical embryonic role[3][2][4][5][6][7].
