Overview

Palmitoyltransferase ZDHHC1 (ZDHHC1) is a human enzyme belonging to the DHHC-type protein acyltransferase family, characterized by a zinc finger DHHC (Asp-His-His-Cys) motif. It catalyzes the reversible S-palmitoylation of a variety of protein substrates, which is a key lipid modification that modulates protein membrane localization, trafficking, stability, and function. ZDHHC1 plays important biological roles in the regulation of antiviral innate immune responses by mediating immune sensor activation as well as in cell growth and tumorigenesis, acting as a potential tumor suppressor in colorectal cancer. It is primarily localized in the Golgi apparatus and endoplasmic reticulum membranes. Downregulation of ZDHHC1 has been linked to unfavorable prognosis in cancer, and genetic defects have been associated with immunodeficiency disorders.
