Overview

Palmitoyltransferase ZDHHC21 is an enzyme of the DHHC-family S-palmitoyltransferases that catalyzes the posttranslational addition of palmitate to substrate proteins, affecting their localization, stability, and function. ZDHHC21 is an integral membrane protein mainly localized to the plasma membrane and the Golgi apparatus. It is known to palmitoylate several key signaling proteins, including GPCRs such as the alpha-1D adrenergic receptor (regulating vascular tone), sex steroid hormone receptors, and the fatty acid synthesis enzyme FASN (acting as a tumor suppressor in DLBCL by destabilizing FASN). Loss of ZDHHC21 function is linked to hypotension and tachycardia due to reduced vascular responsiveness, as well as implications in cancer and neurodevelopmental disorders. ZDHHC21 is considered a potential therapeutic target for modulating vascular tone and certain cancers, with lanatoside C shown as a compound that can target this enzyme for antitumor effects[1][3][4].
