Overview

Palmitoyltransferase ZDHHC22 is a member of the DHHC family of protein palmitoyltransferases, characterized by the presence of a zinc finger DHHC domain. It catalyzes the S-palmitoylation of target proteins, such as KCNMA1, influencing their localization and function at the plasma membrane. ZDHHC22 has enzymatic activity dependent on its DHHC motif and can regulate key signaling pathways by palmitoylating substrates like mTOR, thereby affecting the AKT/mTOR pathway. In breast cancer, ZDHHC22 acts as a tumor suppressor by limiting cell proliferation and promoting apoptosis through cell cycle arrest; its downregulation is associated with increased cancer cell growth and tamoxifen resistance. ZDHHC22 is considered an enzyme target with a potential therapeutic role in oncology, though no direct drug modulators or inhibitors are currently available or reported in clinical or preclinical studies.
