Pancreatic ductal epithelial cell differentiation refers to the process by which progenitor or stem-like cells within the pancreatic ductal epithelium develop into mature, specialized ductal epithelial cells. This process is crucial for normal pancreas development, maintenance of tissue homeostasis, and regeneration after injury. It also plays a role in disease contexts such as diabetes and pancreatic cancer. The differentiation process involves complex signaling pathways (e.g., Notch) that regulate whether a progenitor remains undifferentiated or commits to a specific lineage. Disruption leads to developmental defects or contributes to diseases. Ducts may serve as reservoirs for regeneration/replacement of lost cell types. Experimental models show duct-derived cells can transdifferentiate toward β-cell-like phenotypes under specific protocols mimicking developmental cues.
