Overview

PANK1 antisense RNA 1 (PANK1-AS1) is a long non-coding RNA (lncRNA) that is transcribed from the antisense strand at the PANK1 gene locus. Antisense RNAs such as PANK1-AS1 typically regulate the expression of their corresponding sense genes—in this case, pantothenate kinase 1 (PANK1)—either by interfering with transcription, modifying chromatin structure, or affecting the stability of sense mRNA transcripts[3][6]. While the general function of antisense RNAs is to modulate gene expression at transcriptional and post-transcriptional levels, specific biological roles for PANK1-AS1 have not been well characterized in the current literature. There is thus insufficient evidence that PANK1-AS1 itself functions as a direct therapeutic target such as a receptor, enzyme, or transporter. Instead, it likely exerts regulatory effects, particularly in processes involving gene expression control and possibly epigenetic regulation[3][6]. Antisense RNAs are of interest in disease settings, especially cancer, due to their roles in gene regulation, but direct disease associations and pharmacological targeting of PANK1-AS1 have not yet been reported. Note: - This entry is distinct from PANK1 (pantothenate kinase 1), the protein-coding gene/enzyme, which is a therapeutic target involved in CoA biosynthesis[4][8][10]. - No drugs or biomarkers are currently associated with PANK1-AS1 specifically. - There are no established safety concerns because it is not a direct therapeutic target; however, manipulation of lncRNAs can have broad effects given their key regulatory functions[3][6].
