Pantothenate Kinase (PanK)

Target
PanK
Molecular classification
Enzyme, Kinase
01

Overview

Pantothenate kinase (PanK), also known as CoaA, is the first and rate-limiting enzyme in the coenzyme A (CoA) biosynthetic pathway. It catalyzes the phosphorylation of pantothenate to 4'-phosphopantothenate, using ATP. Mutations in PANK2 cause pantothenate kinase-associated neurodegeneration (PKAN).

Other names
CoaA
02

Mechanism of action

Phosphorylation of pantothenate (vitamin B5) using ATP as a phosphate donor.

03

Biological functions

Coenzyme A biosynthesisPhosphorylation of pantothenic acidMetabolic regulation
04

Disease associations

Pantothenate kinase-associated neurodegeneration (PKAN)
05

Safety considerations

Targeting CoA biosynthesis could have broad metabolic effects.
