Pantothenate kinase 1 beta (PANK1β) is a cytosolic enzyme that serves as the rate-limiting step in the de novo biosynthesis of Coenzyme A (CoA), an essential cofactor for energy metabolism and lipid synthesis [1][2]. It is one of the two major splice variants of the PANK1 gene and is widely expressed in tissues such as the liver and kidney, where it regulates CoA levels in response to metabolic demand [3]. PANK1β activity is tightly controlled through feedback inhibition by acetyl-CoA and other long-chain acyl-CoA thioesters [4]. While PANK2 mutations are the primary cause of Pantothenate Kinase-Associated Neurodegeneration (PKAN), PANK1β is a significant therapeutic target because its pharmacological activation can potentially compensate for the loss of PANK2 activity [5]. Small molecule activators like PZ-2891 have been developed to increase PANK1β activity and restore CoA homeostasis in disease models [5].
