The pantothenate kinase pathway refers to the biochemical sequence responsible for the phosphorylation of pantothenate (vitamin B5) as the first and rate-limiting step in coenzyme A (CoA) biosynthesis. The key enzyme, pantothenate kinase (PanK; EC 2.7.1.33), catalyzes this ATP-dependent reaction, converting pantothenate into 4'-phosphopantothenate. This pathway is essential for all living organisms due to CoA’s central role in metabolism. Mutations or dysregulation are linked to neurodegenerative diseases such as PKAN (pantothenate kinase-associated neurodegeneration) and may also play a role in cancer progression.
