Pantothenic Acid-Dependent Enzymes

Molecular classification
Enzyme
01

Overview

Pantothenic acid-dependent enzymes are a group of enzymes that require pantothenic acid (vitamin B5) or its derivatives for their activity. Pantothenic acid is converted into coenzyme A (CoA) and 4'-phosphopantetheine, which are critical cofactors in numerous metabolic pathways involving the metabolism of fatty acids, carbohydrates, amino acids, and other biomolecules. Key examples include Fatty Acid Synthase Complex, Pyruvate Dehydrogenase Complex, and Acetyl-CoA Carboxylase. These enzymes are vital for energy production, lipid metabolism, and biosynthesis of essential molecules. Deficiency in vitamin B5 impairs these processes due to reduced levels or function of these key enzyme systems.

Mechanism of action

Drugs could potentially target the pantothenate kinase enzyme to modulate CoA biosynthesis, thus affecting the activity of downstream pantothenic acid-dependent enzymes. Inhibition of specific CoA-dependent enzymes directly is also a potential mechanism.
