Overview

The parathyroid hormone/parathyroid hormone-related peptide receptor type 1 (PTH1R) is a Class B GPCR that serves as the primary receptor for both parathyroid hormone (PTH) and parathyroid hormone-related peptide (PTHrP). It plays a critical role in regulating skeletal development, bone turnover, and mineral ion homeostasis, particularly calcium and phosphate balance. PTH1R is highly expressed in bone, kidney, and cartilage. Ligand binding activates G proteins, stimulating adenylyl cyclase and increasing intracellular cAMP, or activating the phospholipase C pathway and mobilizing intracellular calcium. Mutations in PTH1R are associated with several skeletal disorders, and it is a therapeutic target for conditions like osteoporosis and hypoparathyroidism.
