The parathyroid hormone receptor 1 (PTH1R) is a class B G protein-coupled receptor that acts as a central regulator of calcium and phosphate homeostasis. It is activated by parathyroid hormone (PTH) and parathyroid hormone-related protein (PTHrP), and primarily signals through Gs proteins to activate adenylyl cyclase and cAMP production. It is highly expressed in bone and kidney, where it plays critical roles in bone resorption and mineral ion reabsorption/excretion. Defects in PTH1R are associated with several skeletal disorders.
