Parathyroid hormone regulation pathway

Molecular classification
Other (biological pathway, not a single molecule or receptor)

Overview

The "Parathyroid hormone regulation pathway" is not a single molecule or receptor, but rather refers to the complex physiological process by which the secretion and action of parathyroid hormone (PTH) are controlled in the body. PTH is secreted by the parathyroid glands primarily in response to low serum calcium levels. The main regulator is extracellular ionized calcium, sensed by calcium-sensing receptors on parathyroid cells; low calcium stimulates PTH release while high calcium inhibits it through negative feedback mechanisms. Magnesium also plays a role in modulating this secretion. The released PTH acts on bones (stimulating bone resorption), kidneys (increasing reabsorption of calcium and excretion of phosphate), and indirectly increases intestinal absorption of both minerals via activation of vitamin D synthesis[1][2][3]. Dysregulation can lead to various diseases such as hyperparathyroidism, hypoparathyroidism, osteoporosis, and rickets. Note: This entry does not correspond to a specific druggable target such as a receptor or enzyme but rather describes an entire regulatory biological pathway. Therefore, it should be flagged as incorrect for use where only individual molecular targets are appropriate.
