Overview

Parathyroid hormone-related protein (PTHrP) is a proteinaceous hormone and a member of the parathyroid hormone family. It is secreted by mesenchymal stem cells and, in certain pathological conditions, by cancer cells. PTHrP primarily acts via the parathyroid hormone type 1 receptor (PTH1R), regulating bone development, calcium homeostasis, smooth muscle relaxation, and cellular proliferation/differentiation. Overproduction can occur in malignancies leading to humoral hypercalcemia of malignancy. Blood tests measuring circulating levels are used primarily when evaluating unexplained hypercalcemia suspected to be related to malignancy.
