Overview

Parietaria-specific CD4+ T-cell receptors are specialized antigen receptors expressed on the surface of CD4+ T lymphocytes that recognize allergenic peptides derived from Parietaria pollen, such as Par j 1 and Par j 2, presented by Human Leukocyte Antigen (HLA) class II molecules (PubMed: 12642864). These receptors play a central role in the pathogenesis of Parietaria-induced allergic diseases, including allergic rhinitis and asthma, by initiating a Th2-polarized immune response upon exposure to the pollen (PubMed: 10851111). In allergic individuals, the interaction between these TCRs and the peptide-HLA complex triggers the release of pro-inflammatory cytokines like IL-4 and IL-13, which drive IgE production and eosinophilic inflammation (PubMed: 9101110). Therapeutic strategies targeting these receptors primarily involve allergen-specific immunotherapy (ASIT), which aims to induce peripheral T-cell tolerance or shift the immune response toward a regulatory or Th1 phenotype (PubMed: 15606302). Understanding the specific TCR repertoire and the peptide-HLA binding motifs is crucial for developing precision vaccines and monitoring the efficacy of desensitization treatments. These TCRs are often characterized by specific V-beta chain usage and are restricted by specific HLA alleles, most notably HLA-DRB1*11:01 in Mediterranean populations (PubMed: 9101110).
