PACRG (Parkin coregulated gene protein) is an evolutionarily conserved protein encoded by the PACRG gene, closely co-regulated with the adjacent PRKN (Parkin) gene via a bidirectional promoter. It forms large molecular complexes with heat shock proteins and chaperonin components and localizes to the axonemal doublet microtubules in cilia and flagella, where it is essential for microtubule stability and motility. PACRG plays roles in neuronal survival by suppressing cell death triggered by unfolded Pael receptor and is a component of Lewy bodies found in Parkinson's disease brains. It also facilitates TNF-induced NF-κB signaling by stabilizing the LUBAC complex, substituting for the adaptor protein SHARPIN and protecting cells from apoptosis triggered by TNF. Genetic variations in PACRG, especially in its regulatory regions, are linked to increased susceptibility to leprosy and possibly other intracellular infections. There are currently no known drugs directly targeting PACRG, and its role as a therapeutic target has not been established in clinical practice.
