PARP1 binding protein (PARPBP) is a nuclear protein that directly interacts with Poly(ADP-ribose) polymerase 1 (PARP1) and the proliferating cell nuclear antigen (PCNA)[1][4]. PARPBP is part of the DNA replication and repair machinery; it helps maintain genomic stability by protecting replication forks and regulating DNA repair pathways. In cancer, especially hepatocellular carcinoma (HCC), PARPBP is part of the "PCNA/PARP1 axis," where it promotes cell proliferation and therapeutic resistance. Disrupting PARPBP's interaction with PCNA or PARP1 increases cancer cells’ sensitivity to PARP inhibitors (e.g., olaparib), induces apoptosis, and suppresses tumor growth[1]. PARPBP is thus considered a promising therapeutic target, especially in tumors with high DNA repair activity or resistance to existing PARP inhibitor therapies.
Inhibition of PARP1/PARPBP axis increases sensitivity to PARP inhibitors, disrupting compensatory DNA repair pathways. Targeting PARPBP may promote apoptosis and suppress cell proliferation, particularly in cancer cells[1]
