NPR-A (Natriuretic Peptide Receptor A), also known as Guanylyl Cyclase-A (GC-A), is a membrane-bound receptor with intrinsic guanylyl cyclase activity. It is the primary receptor for atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP), playing a crucial role in cardiovascular homeostasis by regulating blood volume and blood pressure through vasodilation, natriuresis, and diuresis. Upon ligand binding, NPR-A dimerizes and catalyzes the conversion of GTP to cGMP, initiating a signaling cascade that affects smooth muscle relaxation, renal function, and cell proliferation.
Ligand binding (ANP, BNP) stimulates guanylyl cyclase activity, increasing intracellular cGMP levels and activating downstream effectors (PKGs, PDEs, CNGs) to mediate vasodilation, natriuresis, and diuresis.
