Overview

PAS domain containing repressor 1 is a transcription factor harboring a PAS domain, evolutionarily related to proteins regulating circadian rhythms. Normally restricted to germline tissues, PASD1 becomes aberrantly expressed in various cancers, where it silences circadian clock activity via direct interaction with the CLOCK:BMAL1 complex, and enhances STAT3-mediated transcription by inhibiting dephosphorylation. PASD1 also suppresses apoptosis and advances cell cycle progression, thus promoting oncogenesis. Because of its restricted normal expression and frequent upregulation in solid and hematological malignancies, PASD1 is being actively investigated as a target for DNA vaccines and T-cell–based immunotherapies in cancer
