Overview

Patched domain-containing protein 3 (PTCHD3) is a member of the Patched family of proteins, featuring a conserved patched domain typical of hedgehog receptors[1][2][5]. It is expressed in male germ cells, particularly on the surface of sperm in mouse, rat, and human[1]. PTCHD3 is evolutionarily conserved, but knockout studies in mice indicate it is not essential for spermatogenesis, sperm physiology, or fertility. Population studies in humans also suggest the gene is non-essential for health or reproduction. While PTCHD3 may be hypothesized to act as a membrane receptor for desert hedgehog (Dhh) or related ligands in spermatogenesis, there is no direct evidence of hedgehog pathway disruption or compensatory biological functions in its absence. As of current knowledge, it is not considered a validated therapeutic target, and no drugs are known to interact with PTCHD3[1][2][5].
