Overview

Patched domain-containing protein 4 (PTCHD4) is a membrane protein belonging to the Patched family, characterized by 12 transmembrane domains and a sterol-sensing domain. It is transcriptionally induced by p53 and regulated via m^6A RNA modification, particularly in response to cellular damage and during senescence. PTCHD4 acts as a putative repressor of the canonical Hedgehog signaling pathway by antagonizing the effects of Smoothened (SMO), demonstrated by the down-regulation of Hedgehog pathway target genes. It sustains the expression of Wnt pathway components relevant to proliferation and survival in senescent fibroblasts and is implicated in the survival of senescent cells and resistance to apoptosis and senolytic agents. Increased PTCHD4 has been observed in cancers such as colorectal and gastric cancer, suggesting roles in tumor biology and cellular aging. No approved drugs specifically target PTCHD4 as of current data.
