Overview

The pathogen membrane is a lipid bilayer structure that forms the boundary of pathogenic microbial cells such as bacteria, fungi, certain parasites, and enveloped viruses. It is typically composed of phospholipids, proteins, and in some organisms, sterols or glycolipids. The membrane confers structural integrity, regulates transport and signaling, and presents pathogen-associated molecular patterns that are recognized by the immune system. Targeting membrane components is a powerful antimicrobial strategy; however, the term "pathogen membrane" is not a precise molecular identifier and encompasses a complex, diverse set of molecules and functions[1][2][3][4][5][6][7].

Mechanism of action

Membrane disruption or permeabilization (by antibiotics/antimicrobial peptides); Inhibition of membrane biosynthesis (e.g., via inhibition of lipid synthesis pathways); Targeting specific membrane proteins (e.g., efflux pump inhibitors); Inhibition of envelope fusion (for enveloped viruses)

05

Safety considerations

Selectivity: Drugs targeting pathogen membranes risk damaging host membranes if not sufficiently selectiveResistance: Bacteria can modify membrane composition to evade drugsToxicity: Non-selective disruption of membranes can cause toxicity to host tissues
06

Interacting drugs

Antimicrobial peptides (e.g., polymyxins, daptomycin)

Biomarkers

Presence of pathogen-specific membrane components (e.g., lipopolysaccharide for Gram-negative bacteria, lipoteichoic acid for Gram-positive bacteria, viral envelope glycoproteins)
