A pathogenic activated CD8 T cell refers to a subset of CD8+ cytotoxic T lymphocytes (CTLs) that, upon activation, acquire effector functions and contribute to disease pathology rather than protection. While typical activated CD8+ T cells are essential for immune defense against infected or malignant cells, the "pathogenic" designation implies these cells are involved in tissue damage or autoimmune responses. These cells kill target cells through the release of cytotoxic granules containing perforin and granzymes, engagement of death receptors such as Fas/FasL pathway, and the secretion of pro-inflammatory cytokines like IFN-γ and TNF-α, which can enhance immune responses but also contribute to inflammation and tissue injury if dysregulated. Pathogenic activated CD8+ T cells are implicated in autoimmune diseases, chronic viral infections, and graft-versus-host disease.
