The pathogenic bacterium cell wall is a complex, mesh-like structure essential for bacterial survival, composed mainly of peptidoglycan (also called murein), which forms a rigid layer around the plasma membrane[1][4][8]. Because peptidoglycan is absent from eukaryotic cells, the bacterial cell wall is an excellent therapeutic target. Beta-lactam antibiotics (such as penicillins and cephalosporins) inhibit enzymes involved in peptidoglycan synthesis, causing cell death[6]. Drugs such as vancomycin, bacitracin, and fosfomycin act on other cell wall synthesis steps. Lysozyme, an enzyme found in human tears and secretions, also degrades peptidoglycan and serves as a natural defense[6]. Significant therapeutic challenges include the emergence of bacterial resistance due to various mechanisms (enzyme production, target modification, efflux), disruption of normal microbial flora, and the risk of massive endotoxin release (e.g., LPS) upon bacteriolysis[6].
