Overview

PATJ crumbs cell polarity complex component (PATJ) is a multi-PDZ domain-containing scaffold protein that is a central member of the apical Crumbs cell polarity complex, which also includes Crumbs (CRB3 in mammals) and PALS1. PATJ localizes to the tight junctions in epithelial cells and is essential for proper apicobasal polarity, organization of tight junctions, and directionality of epithelial cell migration. By serving as an adaptor protein, PATJ links the Crumbs complex to key signaling and structural proteins controlling junction assembly and cellular organization. While disruptions in PATJ or its partners lead to profound defects in epithelial polarity and tissue architecture, PATJ itself is not a current therapeutic target but is critical for understanding epithelial tissue biology and polarity-related diseases.
