Guarana (Paullinia cupana) is a climbing plant native to the Amazon basin, most notable for its seeds which contain a high concentration of caffeine, often twice that found in coffee beans (NIH, 2023). It is not a specific therapeutic target (such as a single receptor or enzyme) but is rather a complex botanical source of multiple bioactive compounds, including methylxanthines and tannins (Wikipedia, 2024). The physiological effects of Guarana are primarily driven by caffeine's antagonism of adenosine receptors, which stimulates the central nervous system and enhances cognitive alertness and physical performance (PubMed, 2019). Additionally, its polyphenol content, specifically catechins and epicatechins, contributes to its antioxidant properties and may play a role in metabolic regulation (PubChem). In the pharmaceutical and nutraceutical industries, Guarana is frequently used in weight-loss supplements and energy drinks, though its complex mixture makes it a source of drugs rather than a target for them.
The primary bioactive constituent of Guarana is caffeine, which acts as a non-selective antagonist of adenosine A1 and A2A receptors in the brain, thereby preventing the inhibitory effects of adenosine and increasing the release of neurotransmitters like dopamine and norepinephrine (StatPearls, 2023). It also inhibits phosphodiesterase enzymes, leading to increased levels of intracellular cAMP, which promotes lipolysis and thermogenesis (PubMed, 2020).
