Overview

PC-esterase domain-containing protein 1A (PCED1A) is a hydrolytic enzyme belonging to the GDSL/SGNH superfamily, characterized by esterase and lipase activity with broad substrate specificity[2][6][8]. It features a catalytic triad of serine, aspartate, and histidine and falls within the Pmr5-Cas1p-esterase subfamily[2][8]. PCED1A is widely expressed and undergoes alternative splicing, resulting in multiple protein isoforms[2][8]. Recent research has identified PCED1A as significantly overexpressed in colorectal cancer (CRC), where it demonstrates strong diagnostic and prognostic utility, and its expression correlates with oncogenic pathways and immune regulation, potentially influencing CRC progression via Notch and Wnt signaling activation[3]. PCED1A may also play roles in chromatin remodeling, immune cell function, and has been suggested as a candidate gene for obesity. While no drugs targeting PCED1A are yet documented, its emerging role as a cancer biomarker makes it an attractive subject for further translational and therapeutic research[3][8].
