PC3-derived tumor-associated antigens represent a collective set of molecular targets found on the surface and within the PC-3 prostate cancer cell line. PC-3 is a classic human cell line derived from a bone metastasis of a high-grade prostate adenocarcinoma, notable for being androgen-independent and lacking expression of Prostate-Specific Antigen (PSA) (ATCC, 2023). In therapeutic contexts, these antigens are utilized in whole-cell vaccines, such as GVAX Prostate, where irradiated PC-3 cells (often combined with LNCaP cells) are used to prime the patient's immune system (Simons & Sacks, 2006). This approach targets a broad array of antigens simultaneously, which helps to address the high degree of clonal heterogeneity found in advanced prostate cancer (Sanda et al., 1994). By presenting these antigens in an inflammatory context—often enhanced by the secretion of Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF)—the therapy aims to break immune tolerance and induce a robust, multi-epitope T-cell attack against the patient's metastatic lesions (van den Eertwegh et al., 2012).
