Overview

PCI domain containing protein 2 (PCID2) is a component of the TREX-2 (Transcription and Export complex 2), involved in nuclear export of mRNA and regulation of cell division checkpoint proteins such as Mad2/MAD2L1. It interacts with and stabilizes BRCA2 protein, controls B-cell survival during differentiation, and restricts lymphoid lineage commitment by modulating chromatin remodeling and gene expression. PCID2 acts as a proto-oncogene in colorectal cancer, promoting cell proliferation, migration, and invasion, while suppressing apoptosis; it mediates these effects primarily by degrading the tumor suppressor PML (promyelocytic leukemia protein), thereby activating canonical Wnt/β-catenin oncogenic signaling and inhibiting noncanonical β-catenin-ARF-p53 tumor-suppressive pathways. High expression or gene amplification of PCID2 is a negative prognostic marker for colorectal cancer recurrence. PCID2 is not currently considered a standard drug target (such as a receptor or enzyme), and there are no reported drugs that specifically interact with or inhibit this protein.
