Overview

PDCD4-antisense RNA 1 (PDCD4-AS1) is a nuclear-enriched long non-coding antisense transcript to the tumor suppressor gene PDCD4. It regulates PDCD4 at the post-transcriptional level by stabilizing its mRNA through direct RNA-RNA interactions and modulation of RNA-binding protein access, notably affecting cell proliferation, migration, and tumorigenesis, especially in breast cancer and gliomas. PDCD4-AS1 is a potential therapeutic and prognostic target; its downregulation is associated with enhanced cancer cell proliferation and poor patient outcomes. PDCD4-AS1 is not a receptor, enzyme, transporter, or classical protein target, but a long non-coding RNA with regulatory functions in cancer biology. It is specifically a NAT, controlling PDCD4 tumor suppressor gene expression through RNA stability mechanisms. Therapeutic targeting has been explored via antisense oligonucleotides in preclinical studies. Acts as an oncogenic factor in certain contexts (e.g., glioma) by regulating miRNA cascades, but generally supports tumor suppression by modulating PDCD4 in breast cancer.
