PDGFA-associated protein 1 (PDAP1) is a ubiquitously expressed, highly conserved, 28 kDa heat- and acid-stable phosphoprotein that modulates growth factor signaling, particularly by enhancing the mitogenic effects of platelet-derived growth factor alpha (PDGFA) on fibroblast proliferation and inhibiting PDGFB-driven growth. Functionally, PDAP1 promotes cell proliferation, migration, and invasion in multiple cellular contexts, notably contributing to the initiation and progression of colorectal cancer via EGFR-MAPK pathway activation. Preclinical and multi-omics studies implicate PDAP1 as a prognostic marker and a potential therapeutic target, with its overexpression correlating with poor survival in cancer patients. Aside from proliferative roles, PDAP1 is involved in immune regulation, supporting mature B cell survival and antibody diversification. No approved drugs currently target PDAP1, but genetic silencing experiments demonstrate its potential utility in cancer therapy.
