Overview

PDGFD downstream neighbor (PDGFDDN, AP002989.1) is a long non-coding RNA gene located on chromosome 11, approximately 150 kb downstream of the platelet-derived growth factor D (PDGFD) gene[1][3]. Genome-wide association studies and functional analyses have shown that variants in this locus may regulate both PDGFD and AP002989.1 expression, influencing smooth muscle proliferation, a key feature in coronary artery disease (CAD) pathogenesis[1][4]. Experimental overexpression of AP002989.1 in human coronary artery smooth muscle cells reduced PDGFD expression and decreased cell proliferation, suggesting a role in vascular biology[1]. However, AP002989.1 is not a protein-coding gene, is not a member of common drug-targetable molecular families (such as receptors, enzymes, or channels), and is not currently considered a direct therapeutic target[1][4].
