Overview

PDZ and LIM domain protein 5 (PDLIM5), also known as Enigma homolog (ENH), is a cytoplasmic protein with a PDZ domain at the N-terminus and three LIM domains at the C-terminus, primarily expressed in heart and skeletal muscle. PDLIM5 acts as a scaffolding protein associated with the actin cytoskeleton, mediating protein-protein interactions important for cytoskeletal architecture, cell adhesion, and signaling. It plays crucial roles in heart and muscle development by scaffolding protein kinase C (PKC) to the Z-disk of cardiomyocytes and regulates cell migration, proliferation, and differentiation. PDLIM5 also sequesters certain nuclear transcription factors in the cytoplasm, thereby modulating gene expression relevant to development and disease. Altered PDLIM5 expression has been implicated in tumorigenesis, metastasis, cardiac hypertrophy, and neuropsychiatric disorders. Multiple transcript variants/isoforms exist with potential tissue- and function-specific roles. However, PDLIM5 is not currently recognized as a direct therapeutic target, and no drugs are known to modulate its function clinically.
