It plays crucial roles in actin filament organization in muscle and non-muscle tissue, regulates transcription factor localization, and participates in both direct and endochondral bone formation, including during fracture repair[4][1]. PDLIM7 also modulates inflammatory responses by acting as an E3 ubiquitin ligase, synergistically facilitating nuclear degradation of NF-κB p65 in cooperation with PDLIM2 and p62/Sqstm1, thus suppressing proinflammatory cytokine production[2]. While it is not an established drug target, its involvement in essential cellular scaffolding, gene regulation, and negative regulation of inflammatory signaling underlies its significance in development, tissue repair, and immune homeostasis[1][2][4][5].
