Overview

PDZ domain-containing protein 11 (PDZD11) is a human protein encoded by the PDZD11 gene. It is a member of the PDZ domain family, acting primarily as an adaptor or scaffold protein. PDZD11 participates in protein-protein interactions at cell-cell junctions, facilitating the docking of ADAM10 to the zonula adherens by interacting with PLEKHA7, which enables PLEKHA7 to associate with the ADAM10-binding protein TSPAN33[2][8]. PDZD11 is involved in pore complex assembly and the regulation of calcium pump (PMCA) localization and calcium handling in cells[2][7][6]. PDZD11 interacts with several proteins, including ATP2B1-4 (plasma membrane calcium ATPases) and ATP7A[2]. While it plays important roles in protein localization and cell-cell junction integrity, there is currently no evidence that it is a direct therapeutic target, nor are there known interacting drugs or established clinical safety concerns. Its gene is located on the X chromosome[6]. Disease associations have been suggested with certain forms of otitis media[2].
