PDZ domain-containing protein GIPC2 is a member of the GIPC family, characterized by a central PDZ domain flanked by unique GH1 and GH2 domains, which enables it to form oligomers and function as an adaptor protein[1]. GIPC2 primarily participates in protein-protein interactions with receptors such as FZD3-type Wnt receptor, insulin-like growth factor receptor, receptor tyrosine kinase A receptor, and TGF-β receptor II, facilitating signal transduction pathways, including those related to Wnt and G-protein[2]. It is implicated in cancer biology, with altered expression or mutations reported in various tumors, such as colon adenocarcinoma, gastric cancer, melanoma, and ovarian cancer[2][3]. Structural studies demonstrate it can oligomerize and act as a scaffold for protein complexes involved in endocytic trafficking and receptor sorting[1]. GIPC2 expression is tissue-specific, highest in the ascending colon and moderate in the kidney[5]. While not currently a direct therapeutic target or known drug receptor, it is recognized for its role in cancer signaling and cell proliferation pathways, and therefore of potential interest in oncology research[2][3].
