PDZ domain-containing protein GIPC3 is a cytoplasmic adaptor/scaffold protein in the GIPC gene family, encoded by the GIPC3 gene. The protein contains a centrally located PDZ domain flanked by GIPC homology domains, enabling it to bind various membrane receptors and signaling proteins. GIPC3 is highly expressed in hair cells of the mammalian cochlea and is critical for proper auditory signal propagation. Mutations in GIPC3 are associated with certain forms of autosomal recessive nonsyndromic hearing loss. While the related GIPC1 and GIPC2 family proteins have broader documented interaction networks, GIPC3 specifically participates in protein complexes via its PDZ domain, modulating receptor trafficking and cellular signaling. There is no current evidence for GIPC3 as a direct therapeutic target or for approved pharmacological agents directed at this protein.
