Overview

PEAK1 related, kinase-activating pseudokinase 1 (PRAG1) is a catalytically inactive member of the tyrosine protein kinase family that functions mainly as a scaffold protein. PRAG1 acts as an effector of the small GTPase RND2, stimulating RhoA activity and inhibiting nerve growth factor (NGF)-induced neurite outgrowth. It also promotes Src family kinase signaling by modulating CSK localization, thus affecting cell morphology and motility. PRAG1 operates as a critical coactivator of Notch signaling. While implicated in various signaling pathways and disease contexts such as cancer and epilepsy, it currently has no known direct drug interactions or established biomarker roles[1][2][7].
