Pediculus humanus capitis, commonly known as the head louse, is an obligate ectoparasite that infests the human scalp and hair (CDC, 2023). It is a wingless insect that feeds exclusively on human blood, requiring frequent meals to survive. While it does not transmit systemic diseases like the body louse, infestation (pediculosis capitis) causes significant scalp pruritus and can lead to secondary bacterial infections such as impetigo due to scratching (StatPearls, 2023). Treatment involves the application of pediculicides, which are chemical agents that target the louse's physiological processes. Most traditional treatments target the insect's nervous system, but the widespread emergence of genetic resistance has necessitated the development of newer agents with different mechanisms, such as metalloproteinase inhibitors or physical suffocants (NCBI, 2020). Effective management often requires multiple applications to ensure both hatched lice and developing embryos are eradicated.
Pediculicides target various physiological systems within the louse: permethrin and pyrethrins disrupt voltage-gated sodium channels (StatPearls, 2023); malathion inhibits acetylcholinesterase (Mayo Clinic, 2023); ivermectin activates glutamate-gated chloride channels (FDA, 2020); and abametapir inhibits metalloproteinases (NCBI, 2020).
