Overview

Peptidase M20 domain-containing protein 1 (PM20D1) is a secreted enzyme encoded by the PM20D1 gene in humans, classified within the mammalian M20 peptidase family[1][2][4]. PM20D1 catalyzes the bidirectional synthesis and hydrolysis of N-acyl amino acids (NAAs) from free fatty acids and amino acids, generating bioactive lipid metabolites that play roles in energy homeostasis, mitochondrial uncoupling, and lipid signaling[1][2][5][8]. It is highly expressed in adipose tissue, pancreas, skin, and widely throughout the brain, circulating in the blood bound to lipoproteins[2][3]. PM20D1 is implicated in metabolic disorders, such as obesity and type 2 diabetes, and neurological disorders, notably as a genetic locus associated with Alzheimer’s disease[1][2]. Its expression is upregulated by the anti-diabetic drug rosiglitazone, and methylation at its locus correlates with body mass index, indicating potential roles as both a therapeutic target and a biomarker[1].
