Overview

The **Peptide YY gene** (PYY) encodes an anorexigenic peptide hormone predominantly produced by enteroendocrine L cells in the gastrointestinal tract, especially the distal ileum and colon, in response to feeding[1][4]. PYY gene expression leads to the secretion of two main peptide forms, PYY 1-36 and PYY 3-36. PYY 3-36 is the major circulating form and acts primarily via the Y2 receptor to inhibit appetite and regulate gastric emptying[1]. Expression of the PYY gene is tightly regulated by dietary nutrients and metabolites such as short-chain fatty acids, which can increase its gene transcription through histone deacetylase inhibition or GPCR pathways (FFA2/GPR43)[2][4]. Alterations in PYY gene expression and hormone levels are implicated in metabolic diseases, including obesity and type II diabetes, with increased interest in leveraging these mechanisms for therapeutic benefit[1][2][3][4]. Peptide YY is also expressed to a lesser extent in skeletal muscle and may have roles in muscle metabolism and regeneration[3]. Direct pharmacological modulation of PYY gene expression is not established as a therapeutic approach; rather, interventions aim to influence its peptide levels or receptor signaling.
