The D-alanyl-D-alanine (D-Ala-D-Ala) terminus is a dipeptide motif found at the end of the peptide side chains in peptidoglycan precursors. It is essential for cross-linking during peptidoglycan biosynthesis and is a target for glycopeptide and β-lactam antibiotics. Resistance can arise from alterations to this terminus.
Vancomycin binds directly to the D-Ala-D-Ala terminus, blocking transglycosylation and transpeptidation. β-lactams mimic the D-Ala-D-Ala terminus and inhibit transpeptidases.
