The peptidoglycan precursor with a D-Ala-D-Ala terminus is a key structural and functional motif in bacterial cell wall biosynthesis. It refers specifically to the terminal dipeptide sequence—D-alanyl-D-alanine—found at the end of the peptide stem attached to N-acetylmuramic acid (MurNAc) residues within peptidoglycan precursors, such as Lipid II. It is essential for cross-linking during cell wall synthesis and is a target for several antibiotics.
Vancomycin binds to D-Ala-D-Ala, blocking transglycosylation and transpeptidation. β-lactams inhibit penicillin-binding proteins.
