Overview

Peptidyl arginine deiminase type-6 (PADI6) is a member of the peptidyl arginine deiminase enzyme family, predominantly expressed in oocytes and early embryos, and is crucial for early embryonic development in mammals[1][2][3]. Unlike other PADIs, PADI6 is catalytically inactive due to the absence of calcium binding and an altered active site, and does not perform the canonical conversion of arginine to citrulline[1][2][3]. Its primary function appears structural, as an essential component of the cytoplasmic lattices needed for the storage and regulation of maternal ribosomes and proteins that govern epigenetic reprogramming, translation, and organelle distribution in early embryos[2][3]. Loss or mutation of PADI6 leads to severe female infertility and oocyte/embryo arrest by disrupting these processes[2][3]. No drugs are known to directly target PADI6, and it is not considered a therapeutic target, but gene mutations serve as biomarkers for infertility risk and defective maternal imprinting.
