Overview

Peptidylglycine alpha-amidating monooxygenase (PAM) is a bifunctional, membrane-bound enzyme essential for the post-translational modification of peptide hormones and neuropeptides. It catalyzes the conversion of inactive peptidylglycine precursors into bioactive alpha-amidated peptides, a modification critical for the full biological activity, stability, and receptor binding of many signaling peptides. PAM is implicated in various physiological pathways including neuronal communication and hormone action. Dysregulation or mutation can affect multiple systems—potentially contributing to psychiatric disorders or cancer progression due to altered levels of bioactive peptides. Inhibitors targeting PAM’s catalytic domains are under investigation for therapeutic applications such as cancer treatment or modulation of neuroendocrine function.
