Cyclophilins are a family of immunophilins characterized by peptidyl-prolyl cis-trans isomerase (PPIase) activity, crucial for protein folding and chaperone activity. Cyclophilin A (CypA) is the main target of cyclosporin A, mediating immunosuppression by inhibiting calcineurin. Cyclophilins are also involved in viral replication, making them potential antiviral targets.
Cyclosporin A binds to cyclophilin, and the resulting complex inhibits calcineurin, preventing T-cell activation. Inhibition of peptidyl-prolyl cis-trans isomerase (PPIase) activity.
