Peptidylprolyl isomerase A pseudogene 43 (PPIAP43) is a pseudogene transcribed as a long non-coding RNA that shares substantial sequence similarity with the peptidylprolyl isomerase A (PPIA) gene. Unlike protein-coding genes, PPIAP43 does not encode a functional protein but instead produces RNA transcripts that can act as decoys for microRNAs targeting PPIA, thereby indirectly regulating PPIA levels. In the context of small cell lung cancer, transcription of PPIAP43 is upregulated in response to ionizing radiation in radiation-sensitive cell lines, suggesting its value as a biomarker to differentiate responsive from non-responsive tumors. Its primary biological function appears to be RNA-level post-transcriptional regulation rather than acting as a traditional therapeutic target. There are no known drug interactions or direct mechanisms of action attributed to PPIAP43, as it does not produce a protein product nor serve as an enzyme, receptor, or transporter. PPIAP43 itself is not a therapeutic target, but rather a non-coding RNA with potential roles in gene regulation and as a biomarker. The function and disease association of PPIAP43 are generally inferred by its regulatory relationship with its parental gene PPIA (cyclophilin A), which is involved in wider physiological and pathological processes, but the pseudogene is distinct and does not encode a protein. Some information may confuse PPIAP43 with its parental gene PPIA; it is crucial to distinguish these, as PPIA (protein) is a validated drug target, while PPIAP43 (pseudogene/non-coding RNA) is not.
